Salmonella

Salmonella refers to a family of bacteria that causes the most frequently reported food borne illness. The most common types of Salmonella are the serotype S. typhimurium and S. enteritidis. Salmonella are predominantly motile but are non-spore forming. Many isolates exist in two phases a motile phase 1 and a non-motile phase II. The bacteria is gram negative and rod shaped and causes symptoms in humans like vomiting, diarrhea, and cramps especially to the immuno compromised and elderly. Food products where Salmonella is commonly found are chickens, raw meat, eggs, shell fish and in fruits such as water melon and cantaloupe. There are over 2000 serotypes of salmonella and all strains are pathogenic. In order not to have false negatives, the selected test must include all serovars but not show closely related species. Therefore, selecting a good sensitive test is essential. Salmonella can survive for long periods in the environment and water is usually a reservoir. The bacteria can survive freezing but can be destroyed by heat for 10 mins at 75°C.

Conventional Method - Biolife Italiana

Most subspecies of Salmonella can be readily detected by growing them in media containing ferrous sulphate and detecting the production of hydrogen sulphide. However, some species may be atypical. The standard gold method for detecting Salmonella in the lab according to BAM is as follows: Enrichment of 25 g food sample into 225 ml of non-selective enrichment broth, then transferred to RV medium and TT broth, then streaking on BSA, XLD and HE.
