ESCHERICHIA coli 0157

Escherichia coli 0157

E. coli 0157:H7 is a special serotype of E. coli that causes severe illness with haemorrhagic diarrhea and stomach cramps with little or no fever. It can cause addition problems like renal failure and anemia. Other E. coli serotypes may also produce the same symptoms like 0145 or 0104:H4.

Occurance of E. coli 0157

This bacteria can be found in food like under cooked beef, raw milk, unpasteurized juice, vegetables and other raw produce. Many outbreaks have linked to beef products. The occurrence of 0157 isolates in nature is usually low thus making its isolation difficult.

Conventional Method – Biolife Italiana

E. coli 0157 can be cultured using Biolife culture media supplied by Arasains Indonesia. 0157 rapidly ferments lactose as is with other E. coli species. However, 0157 isolates ferments D-sorbitol slowly. Biolife MacConkey agar substitutes lactose with sorbitol and is the medium of choice for isolation of E. coli 057. Colonies in this agar which appear colorless are subsequently tested with 0157 anti-serum or latex agglutination reagents. Tests for e coli are usually carried out with the substrate 4-methylumbelliferyl-B-D-glucuronide (MUG) but 0157 is MUG negative and as such this property is often used as an additional screening test for 0157.

Lateral Flow - RapidChek® E. coli O157

RapidChek® E. coli O157 test system offers an easy to prepare one-step enrichment media, without the need of additional supplements, combined with proprietary antibodies to detect the pathogenic E. coli serotype O157 (incl. H7) in various foodstuff within a minimum of 8 hours. Time to result: 8 - 18 hours (depending on the analyzed sample). Approved by AOAC.

 Benefit:

  • Fast and simple procedure
  • Reliable result
  • Easy resource management

Lateral Flow – VIP® Gold E. coli O157:H7 (EHEC)

Merck Biocontrol Indonesia offer VIP® Gold for EHEC is a single-step visual immunoassay for the detection of E. coli O157:H7 in a variety of foods including dairy products, meats and poultry, fruits and vegetables, nutmeats, seafood, pasta, and liquid eggs using an 18 - 28 h enrichment protocol. It is applicable to detection of E. coli O157:H7 in raw and cooked beef and vegetables using an 8 hour enrichment protocol. Each device contains a proprietary reagent system, which forms a visually apparent antigen-antibody-chromogen complex if E. coli O157:H7 is present. Approved by AOAC OMA.

 Benefit:

  • Fast presumptive screening and easy detection after enrichment steps
  • Direct application of enrichment sample
  • Single result format
  • No skills and no automation required
  • Robust housing format for better handling

ELISA – TRANSIA™ AG (Enzyme ImmunoAssay) EHEC

Confirmation of E. coli 0157:H7 requires the identification of the H7 flagellar antigen Biocontrol Indonesia provides a simpler method, Transia™ AG, for the detection of 0157:H7. This uses a complex proprietary formulation to overcome the challenge of differential bacterial identification. A 96 well microtiter plate allows direct binding with the H7 antigen. Automation is possible with the Gemini Analyzer. Accurate detection is available in just 10 hours for beef and in 20 hours in other foods like poultry, fruit nuts and more. This assay is an AOAC official method.

 Benefit:

  • Accurate and reliable
  • High throughput efficiency – 96 well microtiter plate format
  • Increase Productivity with Automation

Frequently Asked Questions

  • What is the disadvantages for using culture media to detec E coli 0157:H7?

The process is and involves multiple steps and as such increases the chances of making mistakes. Final identification also involves either latex agglutination or other methods of differentiation of the flagellar antigen.

  • Is E coli 0157 a serious problem and should any attention be paid to it?

Yes it is a dangerous bacteria that can cause rapid death in victims. It is very common in meat and it is mandatory for meat companies in USA to test for this organism.

  • Why not just test for E coli and not 0157 as it is so difficult?
