Detection of Shiga Toxigenic E. coli and Shiga Toxin Producing Genes

Shiga toxigenic E. coli refers to a group of non 0157 that produces shiga toxin that causes problems ranging from mild intestinal disease to internal haemorrhage including haemolytic uremic syndrome (HUS). They are also sometimes called verotoxin producing E. coli (VETC) or entero haemorrhagic E. coli (EHEC). E. coli 0157 also produce shiga toxin but a strain of E. coli that is not 0157 but identified as E. coli 0104:H4 caused a large outbreak in Europe in 2011. Other non 0157 E. coli that causes serious illness include 026, 0111 and 0103.

Occurrence of STEC

STEC can be found in the guts of ruminants. The major source that cause disease in humans are cattle but they do not have any symptoms of their presence. Pigs and birds sometimes harbor the bacteria and spread it. E. coli have been frequently detected in uncooked beef, raw milk, unpasteurized juices, red leaf lettuce, alfalfa sprouts, brussels sprouts and contaminated water.

PCR Technology - Assurance® GDS STEC

Merck Biocontrol Indonesia supplies the most sensitive detection of STEC E. coli with the Assurance GDS. The USDA definition of the Top six non 0157 E coli include an immunological as well as genetic component of the specific pathogenicity genes. As such PCR is the only method to definitively identify this bacteria. Therefore, rapid methods that employ a single detection technology will generate a high rate of false positives. This Biocontrol method allows the identification of all TOP STEC E coli including 0157:H7, 026, 045, 0103, 0111, 0121 and 0145. The GDS provides a first level of separation by using the pick pen which is coated with antibodies specific to the TOP STEC O group antigen. This eliminates false positives by separating the target TOP STEC E. coli by their desired O groups. After separation of the Top six STEC plus 0157:H7 the samples are analysed by a single multiplex PCR assay for the presence of eae, stx1 and stx2 genes. Results are differentiated for 0157: H7 and the Top six STEC E. coli. There is only a single enrichment step in mEHEC broth providing a time to result of 10 hours.
